Avuncular DNA tests allow siblings of the alleged father (the niece and/or nephew’s aunts or uncles) to participate in a DNA test to verify their biological relationships with possible family members.When we perform a Avuncular DNA Test, also known as an Aunt/Uncle DNA test, we examine 44 specific DNA markers and then analyze them to determine the statistical probability that a individual is the biological aunt or uncle of a particular child (minor or adult). This testing is often used instead of a paternity or maternity test in determining a direct parental relationship when the father or mother is not available. Since we all inherit our genomic content from our parents who, along with their brothers and sisters, have inherited their genetic material from their parents, our genetic material reflects our relationship with our aunts and uncles.

Unlike a paternity test which will always provide a conclusive 99.99% result, an Avuncular test is different. The laboratory will determine the genetic profile of the alleged relatives and then, based on the type of genetic material exhibited by each person, an kinship index is determined. If the index is less than 1.00, this indicates non-relatedness of the people being tested. If the index is greater than 1.00, this indicates that the two tested individuals are more likely to be true biological relatives. The higher the value of the index, the greater the likelihood that the two individuals are true biological relatives.

Bio-Gene DNA Testing only perform a home Avuncular DNA test using a 44 marker basis. While much higher than the industry standard of 16 markers, we have found that it is in our clients best interests to only perform higher marker testing. You deserve to get the most accurate answers the first time you take a DNA test.

Bio-Gene DNA Testing will not perform 16 marker testing for any Avuncular test, as we do not believe that lower marker tests provide accurate results in most cases. The cost difference in minimal, but the accuracy of the results are much higher using a 44 marker test.

The best case scenario in many cases is to test the aunt or uncle, mother, and child to ensure conclusive results about the father. With the industry standard of 16 markers , when less people are tested more markers are needed, and there is no guarantee of a conclusive result.

We recognize that it’s not always possible to test three or more people and by using a 44 marker basis it is possible to take a statistical approach by testing only the aunt or uncle and child which allows us to derive a conclusive result in almost every avuncular case using only two individuals.

The most accurate method of collecting your DNA sample is to use a buccal swab, which is similar to a “Q-tip” cotton swab, which is rubbed on the inner cheeks of the mouth of those being tested. Sometimes it is not possible to obtain a buccal swab sample. We are able to test alternative samples, normally at only a slight extra cost. The DNA sample should have been stored in a paper envelope or temperature environment for the best possibility of success in extracting the DNA. The sample ideally should not have been stored in a plastic bag for an extended time. When you submit an alternate sample please be aware that, while we have a tremendous success rate in obtaining usable DNA profiles from different sources, there is no guarantee that sufficient DNA will be found on your particular sample.

The Accuracy Of The 44 Marker Avuncular DNA Test

The following information helps to understand the difference between 16 marker test results and 44 marker test results.

Case 1

An alleged aunt, mother and child were tested using the 16 marker test. The results were a probability of relationship at 43.47%, which is too low to be conclusive. In this case, the participants added the mother into the analysis but still ended up with inconclusive results. After testing 44 markers, the probability of relationship increased to 91.43%.

 

Case 2

An alleged uncle and a male child were compared with the 16 marker test and the probability of relationship was found to only be 23.90%. Our laboratory tested the two individuals with the 44 marker test and found that the probability of relationship increased from 23.90% with the 16 maker test to 89.81% with the 41 marker test.

Case 3

In this case, the alleged aunt and child were tested with the 16 marker test and the probability of relationship was only 27.26%. The mother was added and the probability changed to 14.25%. It appeared that the alleged aunt was more likely to be unrelated. We applied the 44 marker test on the three participants and found that the probability of relationship was 96.24%. The dramatic change was due to the fact that the 16 markers were not enough for a conclusive result. As a control, our laboratory also analyzed just the alleged aunt and child with 44 markers without the mother and found the probability of relationship to be 90.24%.

 

The conclusion that can be drawn from these case examples is that a 16 marker DNA test will not produce accurate or conclusive results, in most cases, when performing an avuncular test.

Understanding Your Avuncular DNA Testing Results

Aunt/Uncle DNA testing is based on a Kinship Index as explained above. Below is an example of how your testing report will look. We guarantee the accuracy of our test findings but are never able to guarantee specific results until comparing DNA samples. In this case, the testing results indicate that a brother of the aunt tested against the child IS the biological father. The commonly accepted proof for relationships in performing this type of testing is 80% and greater. In this particular case, had the aunt’s brother not been the father of the child, the conclusion would have indicated that results in place of the probability shown. The reports issued are easy to understand and, most important, conclusive in 99.99% of all testing cases we accept. Using the most accurate testing available we are able to eliminate inconclusive results in almost instance.
