Mitochondrial DNA Test - Maternal Lineage DNA Test

What is included: A mitochondrial analysis between two or more individuals, to confirm your relationship to relatives and possible ancestors through the maternal/female line , USPS Priority shipping of a Journey Genetic Testing collection kit (U.S.A. addresses only), detailed collection instructions, a prepaid overnight return label for your DNA samples (U.S.A. addresses only), lab processing fees, and 100% accurate results from our AABB accredited testing laboratory. Mitochondrial analysis kits can be sent to addresses outside of the United States by USPS First Class International for a small additional fee.

Who is included: Two donors, either female, male, or a combination. Other individuals who want to know if they share the same female lineage (aunts, cousins, grandmothers) may be added at an additional cost as shown on the order form.

How we send the DNA collection kit: USPS Priority mail (U.S.A. only). You will by emailed a tracking number from the USPS when the kit ships. If you are having us send the kit to an address outside of the United States, there is a small additional shipping fee and the kit will ship USPS Priority Mail International.

If you need to send a kit to more than one address: You have the option of sending additional testing kits to other addresses if needed. Each additional kit is $14.95 and includes USPS Priority shipping, collection materials, instructions, and a prepaid overnight UPS return mailer for the samples (U.S.A. addresses). If you wish to send more than one kit please indicate this on the order form. Kits shipped to addresses outside of the U.S.A. pay a small additional shipping charge, and are sent by USPS First Class International.

When your results will be ready: 12-14 business days after the lab receives all of the DNA samples. Each individuals’ mtDNA sequences are compared to see if they share certain regions of the DNA that indicate they come from the same maternal line, and this will be shown on the report. Due to the nature of this type of analysis, we are only able to accept mouth swab DNA samples from one of our DNA collection kits.

Payment Options: We offer several ways to pay when you order a sibling DNA test. You can choose to pay with a credit, debit, or prepaid card with a VISA, MasterCard, Discover, or American Express logo. You may also pay through PayPal, and if you do not already have a PayPal account, you can set one up easily. We also financing options through Affirm. Affirm does not perform a hard credit check. Affirm offers both a Pay In 4 Payment plan, as well as offering up to a 12 month payment plan. Affirm is an independent company that we partner with, however the decision to extend financing is their decision alone.   

Our Mitochondrial DNA Test  will provide answers to the question of whether two or more participants are biologically related through a common maternal (mother's) lineage. This is an indirect way to determine maternity. This test is usually done by adoptees, or those who question their maternal heritage. It is also of benefit to help determine if there may be health related issues that are generational. If the mother is not available, or unwilling, to participate in a DNA test to determine direct maternity, this is the recommended way to prove the relationship. Step-By-Step Directions

The mouth (buccal) swabs are packaged in sterile packaging. You will have two packets. Each packet contains TWO swabs. You will use all 4 swabs for each person.

Step 1: Complete the information on the DNA collection envelopes.

The names listed should be the same as you put on your order form. There will be one collection envelope for each person whose DNA is being collected.

Step 2: Open the paper packaging and remove the swabs.

Please be careful to handle the swabs on the long handle (the non-cotton end) only. DO NOT, at any time, touch the cotton end with your hands or fingers.

Step 3: With clean dry hands, collect cheek cells by rolling the tip of the sample collection swab firmly on the inside of the right cheek and under the tongue for 20-30 seconds.

Be sure to roll the swab over the entire inner cheek surface. Repeat this process for the same individual with the second swab. You should use two swabs on the inside of the right cheek and two swabs on the inside of the left cheek. Repeat the process for every individual being tested using a different set of swabs and envelope for each person.

Step 4: The mouth swabs should be dry before putting them into the collection envelope.

The best way of doing this without risking contamination is to place them in an upright position in a clean dry glass. Do not put more than one person's DNA samples into a glass at a time. The DNA samples of each person must never touch each other. Once they are dry, place them back into the correct envelopes.

Step 5: Place all of the swab envelopes, plus and any included forms that you have completed, into the large pre-paid FedEx return envelope.

Then simply return them to a FedEx office or call FedEx to pick it up at a time that fits your schedule.

What’s (Mitochondrial) mtDNA?

Mitochondrial DNA (mtDNA), found within mitochondria, stands apart from nuclear DNA by its exclusive maternal origin. Unlike nuclear DNA, which is a blend of parental DNA, each person's mtDNA closely mirrors that of their mother. With a limited number of genes (37 in total), mtDNA contrasts with the nucleus, which contains approximately 20,000 genes.

In contrast to the two copies of each gene in the nucleus, mitochondria can house 10 or more copies of each gene, and with hundreds to thousands of mitochondria in every cell, the total copies of each mitochondrial gene become substantial.

The unique attributes of mtDNA render it valuable for specific DNA testing applications. In crime scene investigations, the quantity of mtDNA plays a crucial role in determining the information forensic scientists can extract and whether they can identify individuals present at the scene. In cases where nuclear DNA is insufficient for analysis, mtDNA often proves adequate due to its abundance in every cell and its greater stability compared to nuclear DNA. While mtDNA analysis may not always pinpoint a specific individual, it can identify a particular family, making it a useful tool in forensic contexts.

When Should I Consider A Mitochondrial Test?

Here is an example. Consider a scenario where a female Child "A" and her alleged half-brother Child "B" (with the possibility of having the same or a different father than Child "A) seek to determine if they share the same biological mother or maternal grandmother. If the mtDNA test results reveal an identical mtDNA profile, it strongly suggests that they belong to the same maternal lineage. Conversely, if the mtDNA profiles do not match, it indicates that they do not share the same maternal lineage. This highlights the utility of mtDNA testing in establishing maternal relationships between individuals and clarifying their familial connections.

Can Mitochondrial DNA Testing to Determine Ancestry

An extensively employed application of mitochondrial DNA (mtDNA) in DNA testing is the determination of ancestry. Due to its relatively slower rate of change compared to nuclear DNA and its exclusive maternal lineage, mtDNA offers a clearer historical record, specifically of maternal ancestry. This is attributed to the fact that it remains unaltered by paternal DNA.

The analysis of mtDNA has played a pivotal role in allowing scientists to trace the maternal ancestry of the entire human population back to East Africa, with the identified time frame falling approximately between 100,000 and 200,000 years ago. In contrast to regular DNA testing, mtDNA testing can reveal certain aspects of ancestry that may not be readily apparent.

For individuals without a scientific background, mtDNA serves as compelling evidence of relationships among those sharing the same maternal lineage. The shared nature of mtDNA among individuals with common maternal ancestry provides clear confirmation of connections extending from mothers to grandmothers, great-grandmothers, and so forth.

What Else Does MtDNA Help Me Learn?

Mitochondrial DNA (mtDNA) testing holds significant importance in diagnosing certain diseases, particularly those caused by mutations in mtDNA genes. These mutations have been linked to a variety of conditions, often manifesting as neuromuscular diseases or disorders resulting in muscle weakness. A notable case highlighting the impact of mtDNA mutations is that of Greg Lemond, a three-time Tour de France winner, who abandoned competitive cycling due to muscle weakness and fatigue. In 1994, Lemond was diagnosed with "mitochondrial myopathy," bringing mitochondrial diseases to public attention through his story featured in The New York Times.

Scientists have also identified associations between mutations in mtDNA and symptoms of aging-related diseases. This revelation has led to the hypothesis that changes in mitochondrial DNA play a crucial role in determining the pace of aging in individuals. In the future, mtDNA testing may emerge as a key component in assessing overall health. Even subtle alterations in mtDNA could hold significance in gauging an individual's energy levels and overall well-being.

Everyone Lives In Different Locations. Can I Still Do The Test?

In cases where test participants reside at a distance, making it impractical to gather for DNA collection, we provide a convenient solution. We offer the option to ship multiple collection kits to different addresses. When placing your order, you can choose the additional DNA kit option.

To ensure accuracy and proper sample tracking, our collection kits are connected through Journey Genetic Testing case numbers and are matched in lab. The unique case numbers serve as identifiers, allowing us to easi;y match all samples to your specific case. This process ensures reliability and precision in our DNA testing services, even in situations where participants are geographically separated.

Your DNA test results will be easy to read and understand. We will also provide you with our "Understanding Your DNA Results" booklet.

We are providing examples of a Mitochondrial Test report below to give you an idea of what the results you receive will look like. 

There are only two possible results on this type of relationship/kinship DNA test. If the relationship is confirmed, the report will show that the relationship is clearly supported. If there is not a maternal line relationship, then it will also indicate that. In the example below, the relationship does NOT exist.
